“To me, it is now a question of sovereignty,” she said to me last week. “I will personally establish a Planned Parenthood clinic on my own land which is within the boundaries of the Pine Ridge Reservation where the State of South Dakota has absolutely no jurisdiction.”

The reservation is (technically) soverign land, separate from the State of South Dakota and the good ol' US of A.
However...(you knew there was a "however", didn't you?) If the US Government decides to get pissy, they can withhold the federal aid given to reservations until the clinic is closed. Also, the State may decide that assistance to the reservation during a time of need (such as a range/wildfire)isn't within their means - as long as the clinic stands. Indeed, everything from electrical service to water/sewer/trash can be withheld by the State as a means to make the reservation comply.
Of course, the potential PR fallout means that neither of these scenarios are likely to occur, but you never know. When the potential to abuse power exists, power will be abused.
